亚低温对复苏后大鼠脑细胞凋亡相关因子的影响
Effect of mild hypothermia on the apoptosis-related factors in rats after resuscitation

摘要(Abstract):

目的 研究亚低温对心肺复苏后大鼠神经元凋亡及凋亡基因表达的影响。方法 60只SD大鼠随机分为常温(T=37±0.5℃)和亚低温(T=33±1.0℃)组,窒息法建立心肺复苏模型,于自主循环恢复(ROSC)后12h、24h观测神经功能及脑细胞凋亡情况,并于ROCS后0h、2h、6h、12h、24h检测脑组织中AIF、caspase-3、Fas的mRNA基因的表达。结果 亚低温组大鼠12h及24h神经元凋亡减少,神经功能缺损评分偏高;各时间点AIF、caspase-3、Fas的mRNA基因表达显著下调。结论 亚低温可通过抑制脑组织中AIF、caspase-3、Fas的mRNA凋亡基因表达减少神经元凋亡,改善神经功能。
Objective To study role of mild hypothermia therapy after resuscitation on rat neuronal apoptosis and its gene expression, and further explore the mechanism of mild hypothermia treatment on cerebral ischemia-reperfusion injury. Methods 60 healthy male SD rats were randomly divided into 2 groups at normal temperature (T=37±0.5℃) and mild hypothermia (T=33±1.0℃), then were restoration of spontaneous circulation (ROSC) 0h, 2h, 6h, 12h, 24h, divided into 10 subgroups (n=6). Using an asphyxial CA model, we observed the rat nerve function (NDS) at 12h, 24h, TUNEL staining rat brain cell apoptosis, using RT-PCR techniques detected AIF、caspase-3、Fas mRNA gene expression levels of rat brain on 0h, 2h, 6h, 12h, 24h after ROSC. Results Hypothermia rats 12h and 24h neurological deficit scores were higher, TUNEL results showed that the number of 12h and 24h brain cell apoptosis decreased, AIF、caspase-3、Fas mRNA gene expression was significantly reduced at each time point in mild hypothermia group. Conclusion Therapeutic hypothermia after resuscitation can improve neurologic deficit score (NDS), play a role of brain protection, through inhibiting AIF、caspase-3、Fas mRNA of apoptotic genes expression in the rat brain tissue.
